What Is Weight Loss and Cancer Risk?

Obesity is a leading modifiable risk factor for at least 13 cancers — including breast (postmenopausal), colorectal, endometrial, kidney, liver, esophageal, pancreatic, and ovarian. Sustained weight loss of 5 to 10% of body weight significantly reduces cancer risk. Emerging data on GLP-1 medications suggests they may also independently reduce certain cancer risks.
How obesity drives cancer risk
Excess adipose tissue increases chronic inflammation, raises insulin and IGF-1 levels, alters sex hormone balance (especially estrogen), and disrupts immune surveillance — all mechanisms linked to cancer initiation and progression.
Cancers most reduced by weight loss
Breast cancer (postmenopausal) risk decreases significantly with sustained loss. Endometrial cancer risk drops substantially. Colorectal, kidney, and liver cancer risks also decline. The earlier and more sustained the weight loss, the greater the benefit.
Emerging GLP-1 data
Observational studies suggest semaglutide and tirzepatide may reduce risk of obesity-associated cancers independent of weight loss — possibly through anti-inflammatory and metabolic effects. Randomized data is still pending.
